How to Make Mushroom Liquid Culture

You can use this recipe to learn how to manufacture oyster mushroom liquid culture. In actuality, this recipe works with any type of mushroom.


What you will need:

  • Grain spawn jar, also known as an airport jar
  • Pressure cooker
  • 10ml syringes
  • Needles (Note: the bigger the number, the smaller the needle. I recommend 18 gauge)
  • One of the following sugars to mix with water: organic honey, corn syrup, corn sugar, light malt extract, or dextrose (glucose).

Note: Sucrose (household sugar) should not be used. For this guide, honey was chosen.


What is the best sugar-to-water ratio?

The ideal ratio is 4%. For every 100 ml of water, this works out to 4cc, 4ml, or 1 teaspoon of honey. The ratio should be between 3% and 5%, but too much sugar (more than 10%) will kill the mycelium.


A step-by-step guide to making your own mushroom liquid culture:

  • Carefully measure (using eco-friendly mearing cups, of course!) and mix the sugar you choose with water in the jar. Warm water will help the sugar dissolve quickly, but it is optional.
  • Put the lid on the jar.
  • Cover the top of the jar with aluminum foil and put it in a pressure cooker.
  • Cook at 15 psi for no longer than 15-20 minutes. Over-cooking the sugar will lead to caramelization, resulting in poor to no mycelium growth.
  • Allow the pressure cooker to cool off naturally before removing the jar. Be careful. The jar will be very hot.
  • Allow the jar and contents to reach room temperature.
  • Add some mother culture to your liquid culture.

Note: If you are not using an airport jar, keep the lid on until you are prepared to inoculate your substrate and put it back on as soon as you finish. By doing this, airborne contaminants will not get inside the jar and harm your mycelium.

How to add mother culture to your liquid culture

You can add a few ccs of mother culture after the jars have cooled after being in the pressure cooker. Break apart the mycelium by vigorously shaking the mother culture.

Use an environmentally friendly soap to thoroughly wash and dry your hands with a neat paper towel. A syringe needle being used for the first time should naturally come sterilized. If not, hold the needle in the flame of a candle or lighter until it is red hot to sterilize it. Be careful now! To cool the needle down, clean it with an alcohol swab.
